About the Project

Intracellular Organization Dynamics is an online resource that contains information about the morphological dynamics of 21 organelles and major subcellular structures upon conditional inhibition of 352 essential genes and progressive transition towards cell death (Litsios et al. 2026). To retrieve this information, individual organelles and subcellular structures were visualized in Saccharomyces cerevisiae (budding yeast) via the fusion of native proteins with GFP. Conditional gene inhibition was achieved via the use of temperature-sensitive alleles. Live cells were monitored every 3 hours following a shift to restrictive temperature during a 24-hour time course using automated confocal microscopy. The resource contains both the raw images of the project, as well as data that were extracted from their analysis.

How to use the resource

At the main page of the project (https://thecellvision.org/terminalphenotypes), the user types or selects their gene/mutant of interest. Here, either Yeast gene names or the names of their Human orthologues can be used, and multiple genes can be queried at the same time. The user is then directed to the respective gene(s) page, where images of the mutant(s) – along with wild type controls – are displayed for each of the visualized organelles and subcellular structures and each of the nine time points. The quantified percentage of cells with abnormal morphology in a respective structure (penetrance) at each time point is also displayed in tabular format. Both the raw images and the analyzed penetrance data can be downloaded via the respective prompts.
